以牛的基因组DNA为材料,对影响牛SRAP-PCR扩增的各因素浓度进行了优化,建立了重复性好、稳定性高的SRAP-PCR反应体系。结果表明在30μL的SRAP反应体系中5种成分的最适浓度为:Mg~(2+) 1.6 mmol/L、模板DNA 60 ng、引物0.20μmol/L、dNTPs 60μmol/L、Taq DNA聚合酶1.0 U。
Using the genomic DNA of cattle as material, the concentration of each factor influencing the amplification of SRAP-PCR in cattle was optimized, and the SRAP-PCR reaction system with good repeatability and high stability was established. The results showed that the optimal concentrations of the five components in the 30μL SRAP reaction system were: Mg 2+ 1.6mmol / L, template DNA 60ng, primer 0.20μmol / L, dNTPs 60μmol / L, Taq DNA polymerase 1.0 U.
