目的:建立HPLC测定芒萁中槲皮素和山柰素含量的方法,并测定不同产地芒萁中槲皮素和山柰素含量。方法:分别用不同体积分数的甲醇和乙醇做为槲皮素和山柰素的提取溶剂,采用超声、回流两种方法提取槲皮素和山柰素,采用高效液相色谱法,Diamonsil C18色谱柱(4.6 mm×200 mm,5μm),流动相甲醇-0.1%磷酸水(54∶46),流速1.0 m L·min-1,柱温30℃,检测波长366 nm。结果:槲皮素和山柰素用80%甲醇回流提取率较高,江西鹰潭的芒萁中槲皮素和山柰素含量较高。芒萁中槲皮素、山柰素分离效果良好,线性范围分别为1.051~16.24,0.505~8.08μg。结论:该法准确灵敏、稳定可靠,可用于芒萁的质量控制,不同产地芒萁中槲皮素及山柰素含量差别较大。
OBJECTIVE: To establish a HPLC method for the determination of quercetin and kaempferol in mangosteen and to determine the contents of quercetin and kaempferol in mangosteen from different habitats. METHODS: Methanol and ethanol with different volume fractions were used as extraction solvent for quercetin and kaempferol respectively. Quercetin and kaempferol were extracted by ultrasonic and reflux methods. High performance liquid chromatography (HPLC) and Diamonsil C18 The mobile phase was methanol-0.1% phosphoric acid water (54:46) at a flow rate of 1.0 mL · min-1. The column temperature was 30 ℃ and the detection wavelength was 366 nm. Results: The extraction rate of quercetin and kaempferol with 80% methanol was higher than that of quercetin and kaempferol in Yingtan of Jiangxi Province. Manglietia quercetin, Shan Su Su isolated good results, the linear range was 1.051 ~ 16.24,0.505 ~ 8.08μg. Conclusion: The method is accurate, stable and reliable and can be used for the quality control of Mangosteen. The contents of quercetin and kaempferol in Mangosteen are different from each other.
