目的 探讨肿瘤坏死因子 α(tumornecrosisfactoralpha ,TNF α)对周围神经损伤后感觉神经元的保护作用。方法 取Wistar大鼠 2 0只 ,分成TNF α和SAL两组。右侧坐骨神经切断后 ,将神经近断端套入硅胶管内。TNF α组自硅胶管内注入 16μlTNF α( 3 0U/ml)。SAL组在硅胶管内注射等量生理盐水。术后 2周观察脊神经节内感觉神经元乙酰胆碱脂酶 (AChE)、一氧化氮合酶 (NOS)的变化 ;测量脊神经节神经元的存活百分率及乙酰胆碱酯酶的平均灰度。结果 两组的乙酰胆碱脂酶、一氧化氮合酶均较正常神经元有显著性的变化 ,两组间神经元的存活百分率及乙酰胆碱酯酶的平均灰度的差异也存在非常显著性意义 (P <0 .0 1)。结论 TNF α对周围神经损伤后脊神经节感觉神经元有保护作用。
Objective To investigate the protective effect of tumor necrosis factor alpha (TNF α) on sensory neurons after peripheral nerve injury. Methods Twenty Wistar rats were divided into two groups: TNF α and SAL. After the right sciatic nerve was dissected, the proximal nerve end was placed in a silicone tube. In the TNF α group, 16 μl of TNF α (30 U/ml) was injected into the silicone tube. The SAL group was injected with equal volume of saline in a silicone tube. The changes of acetylcholinesterase (AChE) and nitric oxide synthase (NOS) in the sensory neurons of the dorsal root ganglion were observed 2 weeks after surgery. The percentage of survival of spinal ganglion neurons and the average gray level of acetylcholinesterase were measured. Results The levels of acetylcholinesterase and nitric oxide synthase in the two groups were significantly different from those in normal neurons. There was also significant difference in the survival rate of neurons and the mean gray level of acetylcholinesterase between the two groups. <0. 0 1). Conclusion TNF α has a protective effect on spinal ganglion sensory neurons after peripheral nerve injury.
