利用体外培养技术,以K562细胞为实验模型,通过K562细胞的生长曲线、细胞形态、乳酸脱氢酶活性、血红蛋白测定等,观察了苦参提取液对K562细胞的诱导分化作用。以MTT法结合细胞形态探讨苦参提取液作用K562细胞的有效分化浓度及有效作用时间,研究结果表明:12mg/ml浓度的苦参提取液作用K562细胞二天,从形态到代谢都发生了变化。LDH活性明显降低;血红蛋白生成;细胞增殖明显受到抑制。电镜形态提示,细胞向似红细胞系、粒细胞系及单核细胞系分化。
Using in vitro culture technology, K562 cells as an experimental model, through the growth curve of K562 cells, cell morphology, lactate dehydrogenase activity, hemoglobin determination, etc., observed the effect of Sophora on the differentiation of K562 cells. Using MTT assay combined with cell morphology to explore the effective differentiation concentration and effective duration of K562 cells treated with Sophora flavescens extract, the results showed that: Sophora flavescens extract with 12mg/ml concentration acted on K562 cells for two days and changed from morphology to metabolism. LDH activity was significantly reduced; hemoglobin was formed; cell proliferation was significantly inhibited. The morphology of the electron microscope suggests that the cells differentiate into erythroid, granulocyte, and monocyte lines.
