抵抗素(resistin)自2001年被发现以来,人们对其生物学功能尚不清楚。本研究目的是通过抵抗素的组织表达分布以探讨人抵抗素的生物学功能、表达及与白血病发病的相关性。选择152例无炎症并发症的白血病病人作为实验组,100例健康志愿者作为对照组,采集静脉血血样,采用半定量PCR方法检测抵抗素在组织中的表达分布并比较抵抗素与内参基因表达的相对值,进行统计学分析。结果表明:抵抗素在正常人骨髓、胎儿肝脏,脐带血以及外周血细胞中有高水平表达,而在脂肪、脐带、胎盘以及成人肝脏组织中尚没有扩增出目的基因。实验发现,实验组抵抗素表达率为21%,对照组抵抗素表达率为27%,经卡方检验,χ2=0.84,差异不显著。对实验组与对照组抵抗素表达水平比较结果进行了统计学分析,差别无统计学意义(p>0.05)。结论:抵抗素在正常人骨髓、胎肝、脐血及外周血细胞中有高表达,此结果表明人抵抗素的表达与正常造血相关,但其表达率和表达水平与白血病无明显相关性。
Resistin (resistin) since its discovery in 2001, people are not clear about its biological function. The purpose of this study is to investigate the biological function of human resistin, its expression and the correlation with the pathogenesis of leukemia through the tissue expression and distribution of resistin. Fifty-two patients with leukemia without inflammatory complications were selected as experimental group and 100 healthy volunteers as control group. Venous blood samples were collected. Semi-quantitative PCR was used to detect the distribution of resistin in tissues and to compare the expressions of resistin and internal reference genes The relative value, for statistical analysis. The results showed that resistin was expressed at high levels in normal human bone marrow, fetal liver, umbilical cord blood and peripheral blood cells, but no target gene was amplified in adipose tissue, umbilical cord, placenta and adult liver tissue. The experiment found that the expression rate of resistin in experimental group was 21%, and the expression rate of resistin in control group was 27%. The chi-square test showed that χ2 = 0.84, the difference was not significant. The experimental group and control group resistin expression levels were compared between the statistical analysis, the difference was not statistically significant (p> 0.05). Conclusion: Resistin is highly expressed in normal human bone marrow, fetal liver, umbilical cord blood and peripheral blood cells. The results indicate that the expression of human resistin is related to normal hematopoiesis, but the expression and expression level of resistin is not significantly correlated with leukemia.
