[目的]观察PIAS3在结肠癌组织及细胞中的表达,探讨PIAS3在结肠癌发生发展中的作用。[方法]采用免疫组织化学染色法检测60例结肠癌及同源癌旁组织中PIAS3蛋白的表达,实时荧光定量PCR(q RT-PCR)、Western blot检测人结肠癌SW480、SW620及COLO205细胞株中PIAS3 m RNA及蛋白表达水平。[结果 ]结肠癌组织中PIAS3蛋白表达显著高于癌旁正常组织(χ~2=8.543,P=0.006);且表达强度随肿瘤病理分化程度的降低、Dukes分期的增高及肿瘤体积的增大而逐渐增高,差异有统计学意义(P均<0.05);PIAS3在结肠癌SW480、SW620及COLO205细胞株中均有表达,其中COLO205细胞株中PIAS3蛋白及m RNA表达高于SW480和SW620细胞株(P<0.05)。[结论 ]PIAS3的表达水平以及活性与结肠癌的发生发展有关,可能为结肠癌的早期诊断及靶向治疗提供新的分子靶点。
[Objective] To observe the expression of PIAS3 in colon cancer tissues and cells and explore the role of PIAS3 in the development of colon cancer. [Methods] The expression of PIAS3 protein was detected by immunohistochemical staining in 60 cases of colorectal cancer and its adjacent tissues. Real-time quantitative PCR (q RT-PCR) and Western blot were used to detect the expression of PIAS3 protein in human colon cancer SW480, SW620 and COLO205 cell lines PIAS3 m RNA and protein expression level. [Results] The expression of PIAS3 protein in colon cancer tissues was significantly higher than that in adjacent normal tissues (χ ~ 2 = 8.543, P = 0.006). The expression intensity of PIAS3 was decreased with the differentiation of tumor, Dukes stage and tumor volume (P <0.05). PIAS3 was expressed in SW480, SW620 and COLO205 cell lines, and the expression of PIAS3 protein and m RNA in COLO205 cells was higher than that in SW480 and SW620 cell lines (P <0.05). [Conclusion] The expression level and activity of PIAS3 are related to the development of colon cancer, which may provide a new molecular target for the early diagnosis and targeted therapy of colon cancer.
