目的通过检测和比较子宫内膜异位症(EMS)孕妇有无补充叶酸其女性胎儿脐血EMS易感基因HOXA10启动区CpG岛甲基化状态,探讨叶酸优化宫内环境的作用。方法收集标本2010年1月~2012年12月21例患有EMS孕妇服用和15例未服用叶酸的女性胎儿脐血标本,利用甲基化特异性聚合酶链反应(MSP)及亚硫酸氢盐修饰后测序(BSP)检测和比较各组HOXA10的甲基化率。结果补充叶酸组女性胎儿EMS易感基因HOXA10甲基化率显著降低,两组差异有统计学意义(P<0.05)。结论提示叶酸在预防胎源性EMS易感基因HOXA10异常甲基化中可能起一定作用。
Objective To explore the role of folic acid in optimizing the intrauterine environment by detecting and comparing the methylation status of CpG island in the promoter region of HOXA10 promoter in the pregnant women with endometriosis (EMS) with and without folic acid. Methods Twenty-one female cord blood samples from 21 pregnant women with EMS and 15 non-folic acid women were collected from January 2010 to December 2012. Methylation-specific polymerase chain reaction (MSP) and bisulfite Post-sequencing modification (BSP) detected and compared the methylation rate of HOXA10 in each group. Results The methylation rate of HOXA10 in the fetus with folic acid supplementation was significantly decreased (P <0.05). The results suggest that folic acid may play a role in the prevention of fetal methylation of HOXA10 gene.
