目的:观察虎杖苷对海人酸致癫痫鼠颞叶、海马区P-糖蛋白表达的影响。方法:通过脑立体定向技术,将海人酸(KA)1.5μg(3μL)注射至SD大鼠海马,制作颞叶癫痫模型,假手术组为同法于大鼠海马注射3μL生理盐水。将建模成功的26只癫痫大鼠随机分为虎杖苷处理组(n=13)、生理盐水处理组即对照组(n=13)。分别给予虎杖苷(100 mg/kg)、等体积生理盐水灌胃1次/天,均连续30天。观察记录大鼠发作频率、级别及持续时间,并采用免疫组化EnVision法检测大鼠颞叶、海马区P-糖蛋白(P-gp)表达水平。结果:观察期内虎杖苷组在干预后,发作频率及级别、平均持续时间较前明显减少(均P<0.05),而对照组未见差异(均P>0.05),对照组和虎杖苷组P-gp的表达强度均高于假手术组(均P<0.05),与对照组比较,虎杖苷组P-gp的表达水平有所下降(P<0.05)。结论:虎杖苷有一定的抗癫痫作用,下调P-gp的表达可能为其机制之一,为开发新的辅助抗癫痫药物提供思路。
OBJECTIVE: To observe the effects of cedar pollen on the expression of P-glycoprotein in temporal lobe and hippocampus of kainic acid-induced epileptic rats. Methods: The temporal lobe epilepsy model was induced by injection of 1.5μg (3μL) KA into the hippocampus of SD rats by stereotactic technique. The sham operation group was injected with 3μL normal saline into the hippocampus of rats. Twenty-six epilepsy rats with successful modeling were randomly divided into three groups (n = 13) and the saline-treated group (n = 13). Were given cedar glycosides (100 mg / kg), an equal volume of normal saline gavage 1 / day, were 30 consecutive days. The frequency, level and duration of seizure were recorded. The expression of P-glycoprotein (P-gp) in temporal lobe and hippocampus was detected by EnVision immunohistochemistry. Results: During the observation period, the frequency and level of seizure and the mean duration of seizure decreased significantly in the PDG group (all P <0.05), but not in the control group (all P> 0.05) The expression of P-gp was significantly higher in sham operation group than that in sham operation group (all P <0.05). Compared with control group, the expression of P-gp was decreased (P <0.05). Conclusion: Polydatin has antiepileptic effect. Down-regulation of P-gp expression may be one of its mechanisms, providing a new idea for developing new antiepileptic drugs.
