用脾集落技术结合有关方法对小鼠多能造血干细胞(CFU-S)的一些性质与功能做了观察。输入的骨髓细胞数目与所生成的脾集落数目呈线性关系。成年小鼠的造血组织中,CFU-S在骨髓中分布最多,脾脏及外周血中也有一小部分。它在上述器官中的百分比,依次为0.7%、0.05%、0.01%。而胸腺与淋巴结中为阴性所见。在正常状态下,CFU-S大多处于G_0期,S期的比率很低,仅为6.7%。 CFU-S具有重要的自我更新功能,用单个脾集落再移植实验测得其自我更新率P=0.76。CFU-S有主动的迁移性能,当给受致死性照射的小鼠腹腔注入骨髓细胞悬液或游离置入结扎了血管的完整脾脏于其腹腔中时,都可以见到有一部分CFU-S能穿越许多屏障而迁至受体脾脏,形成脾集落。500~700拉德的γ-射线照射,能明显地刺激骨髓CFU-S向血运中迁移。
