目的观察左旋四氢帕马丁(levo-tetrahydropalmatine,L-THP)对大鼠海洛因强化效应和自发活动度的影响。方法雄性SD大鼠14只,用固定比率强化程序建立大鼠海洛因自身给药行为模型,模型建立后动物随机分为2组(n=7):L-THP 2.5mg/kg组和L-THP 5.0mg/kg组,观察腹腔注射不同剂量L-THP对大鼠海洛因固定比率自身给药行为的影响。24只雄性SD大鼠随机分为3组(n=8):溶媒组、L-THP 2.5mg/kg组和L-THP 5.0mg/kg组。观察腹腔注射不同剂量L-THP或等量溶媒对大鼠自发活动度的影响。结果在固定比率程序测试中,L-THP(2.5,5.0mg/kg)可显著抑制大鼠海洛因自身给药行为,与基值相比差异均有统计学意义(P<0.01)。在自发活动度测试中,与溶媒对照组相比,L-THP 2.5mg/kg组活动度无显著性差异(P>0.05),而L-THP 5.0mg/kg组活动度显著降低(P<0.05)。结论 L-THP 2.5mg/kg组可降低大鼠海洛因的强化效应,且不会影响其活动度。
Objective To observe the effects of levo-tetrahydropalmatine (L-THP) on heroin fortification and spontaneous activity in rats. Methods Fourteen male Sprague-Dawley rats were randomly divided into 2 groups (n = 7): L-THP 2.5 mg / kg and L-THP 5.0 mg / kg group. The effects of intraperitoneal injection of different doses of L-THP on self-administration of heroin in rats were observed. 24 male SD rats were randomly divided into 3 groups (n = 8): vehicle group, L-THP 2.5 mg / kg group and L-THP 5.0 mg / kg group. To observe the effect of intraperitoneal injection of different doses of L-THP or equal amount of vehicle on spontaneous activity in rats. Results In the fixed rate program, L-THP (2.5, 5.0 mg / kg) significantly inhibited the self-administration of heroin in rats, with statistical significance (P <0.01). Compared with the vehicle control group, there was no significant difference in activity of L-THP 2.5 mg / kg group (P> 0.05), but the activity of L-THP 5.0 mg / kg group was significantly lower than that of vehicle control group (P < 0.05). Conclusion L-THP 2.5mg / kg group can reduce the strengthening effect of heroin in rats, and will not affect its activity.
