目的:采用高效液相色谱法测定不同产地及不同采收期大花紫薇叶中鞣花酸的含量。方法:采用Ultimate XB-C18(250mm×4.6mm,5μm)色谱柱,柱温25℃,以甲醇-0.1%磷酸水溶液为流动相,梯度洗脱,流速0.8mL·min-1,检测波长254nm。结果:鞣花酸浓度在2.66~66.45μg/mL范围内与峰面积呈良好线性关系,r=0.9998。平均回收率为103.7%(RSD=0.9%,n=6)。不同产地的大花紫薇叶中,鞣花酸以广东深圳采收的含量最高,不同采收时间的福建卫生职业技术学院大花紫薇叶中鞣花酸含量在始果期8月下旬达到最高。结论:本方法操作简单、快速、重复性好,可用于测定大花紫薇叶中鞣花酸的含量。
OBJECTIVE: To determine the content of ellagic acid in leaves of Lagerstroemia indicia from different habitats and harvesting periods by high performance liquid chromatography (HPLC). METHODS: The column was packed with Ultimate XB-C18 (250 mm × 4.6 mm, 5 μm) at a column temperature of 25 ° C using a mobile phase of methanol-0.1% phosphoric acid as mobile phase with a flow rate of 0.8 mL · min-1 and a detection wavelength of 254 nm. Results: The ellagic acid concentration in the range of 2.66 ~ 66.45μg / mL showed a good linear relationship with the peak area, r = 0.9998. The average recovery was 103.7% (RSD = 0.9%, n = 6). In different areas of Lagerstroemia indica, ellagic acid was collected in Shenzhen, Guangdong Province, with the highest content of ellagic acid in different harvest time. The ellagic acid content of Lagerstroemia speciously reached its peak in late August in Fujian Academy of Occupational Health Science and Technology. Conclusion: The method is simple, rapid and reproducible. It can be used to determine the ellagic acid content in Lagerstroemia specious leaves.
