目的探讨唑来膦酸刺激人外周血γδT细胞的增殖作用,同时测定增殖后的γδT细胞对人肝癌细胞的杀伤效能。方法以不同浓度的唑来膦酸联合IL-2刺激健康人外周血γδT细胞增殖。以流式细胞仪检测相关表型,3H-TdR掺入法测定细胞增殖程度。以γδT细胞作为效应细胞,分别以不同人肝癌细胞作为靶细胞。以SRB法测定效应细胞对靶细胞的杀伤效能。结果外周血γδT细胞增殖明显,唑来膦酸为1μmol/L浓度时,γδT细胞增殖最明显。所得γδT细胞对3种肝癌细胞均表现出明显的杀伤作用。结论唑来膦酸能明显促进人外周血γδT细胞的增殖,且扩增的γδT细胞在体外对肝癌细胞有显著的杀伤作用。
Objective To investigate the proliferation of γδT cells stimulated by zoledronic acid and the cytotoxicity of γδT cells on human hepatoma cells. Methods Zoledronic acid combined with IL-2 at different concentrations stimulated the proliferation of γδT cells in healthy human. The related phenotypes were detected by flow cytometry, and the degree of cell proliferation was determined by 3H-TdR incorporation. ΓδT cells as effector cells, respectively, different human hepatocellular carcinoma cells as target cells. The killing effect of effector cells on target cells was determined by SRB method. Results The proliferation of γδT cells in peripheral blood was obvious. When the concentration of zoledronic acid was 1μmol / L, the proliferation of γδT cells was the most obvious. The resulting γδT cells showed obvious killing effects on all three kinds of hepatoma cells. Conclusion Zoledronic acid can significantly promote the proliferation of γδT cells in human peripheral blood, and the expanded γδT cells have a significant killing effect on hepatoma cells in vitro.
