目的:成纤维细胞生长因子(FGF-10)是间质细胞合成与分泌,特异性作用于上皮细胞的生长因子。研究表明 FGF-10在促进动物创面以及人下肢静脉慢性溃疡的愈合过程中,其组织学特征不仅表现在刺激角朊细胞增殖与迁移,还表现在促进肉芽组织的形成。本研究拟通过观察 FGF-10对融合和未融合两种生长状态的角朊细胞表达转化生长因子α(TGFα)的作用来探讨 FGF-10促创面修复的作用机制。方法:FGF-10的工作浓度分为4、16、125和500ng/ml 四组,不含 EGF 和含有 EGF 的无血清角朊细胞培养液分别作为阴性对照组和阳性对照组。细胞接种密度为2500细胞/cm~2或375000细胞/cm~2,分别于 FGF-10作用后24、48和72h 收集细胞培养上清,同时进行细胞计数,上清中 TGFα的含量用 ELISA 试剂盒测定。结果:当细胞接种密度为2500细胞/
AIM: Fibroblast growth factor (FGF-10) is a growth factor that acts on the synthesis and secretion of interstitial cells and specifically on epithelial cells. Studies have shown that the histological features of FGF-10 in promoting the healing of wounds and chronic venous leg ulcers in human are not only in stimulating the proliferation and migration of keratinocytes, but also in promoting the formation of granulation tissue. This study was to investigate the mechanism of FGF-10 in promoting wound healing by observing the effect of FGF-10 on the expression of transforming growth factor-α (TGF-α) in keratinocytes in both growth and non-fusion states.
