目的探讨何首乌饮对过度训练大鼠睾丸组织3β羟基类固醇脱氢酶(3β-HSD)的影响。方法 30只雄性SD大鼠随机分为5组,A组为安静对照组;B组为安静何首乌饮组;C组为自然恢复组;D组为何首乌饮治疗组;E组为何首乌饮预防组,每组6只。C组、D和E组大鼠复制力竭运动动物模型,其中的E组每天训练前给何首乌饮20g/(kg.d)(含生药9.6kg/L)灌胃作为预防。模型成功后D组给何首乌饮20g/(kg.d)(含生药9.6kg/L)灌胃治疗60d。放射免疫法检测各组血清睾酮浓度,分别采用RT-PCR、Western blotting和免疫组织化学法检测各组睾酮合成催化酶3β-HSD的表达变化。结果 3β-HSD蛋白的阳性信号为间质细胞的胞质表达棕黄色颗粒。3β-HSD在组织、蛋白和mRNA的表达变化:B组明显高于A组(P<0.05),E组和D组高于C组(P<0.05),E组和D组之间无显著性差异。结论何首乌饮可以增强睾酮合成限速酶3β-HSD的表达。
Objective To investigate the effects of Heshouwuyin on 3β-hydroxysteroid dehydrogenase (3β-HSD) in the testis in rats. Methods Thirty male Sprague-Dawley rats were randomly divided into 5 groups: group A, a quiet control group; group B, a quiet Ho-Ho-Wu drinking group; group C, a natural recovery group; group D, , 6 in each group. Rats in groups C, D and E were exposed to exhaustive exercise. Animals in group E received intragastric administration of 20g / (kg.d) (9.6kg / L crude drug) before the training in group E as a preventive. Group D after the success of the model given Polygonum 20g / (kg.d) (containing crude drug 9.6kg / L) for 60 days. Serum testosterone levels were determined by radioimmunoassay. The expression of testosterone-synthesizing enzyme 3β-HSD in each group was detected by RT-PCR, Western blotting and immunohistochemistry respectively. Results The positive signal of 3β-HSD protein was expressed in the cytoplasm of stromal cells as brown granules. The expression of 3β-HSD in the tissue, protein and mRNA was significantly higher in group B than in group A (P <0.05), in group E and group D (P <0.05), and in group E and group D Sex differences. Conclusion Polygonum multiflorum Thunb can enhance the expression of 3β-HSD, a rate-limiting enzyme in testosterone synthesis.
