目的观察小鼠泡球蚴感染中期(3月)肝内miR-100、miR-125b、miR-126和miR-122的表达并探讨其意义。方法肉眼直视下穿刺小鼠肝脏,注射0.1ml泡球蚴组织匀浆液,建立泡球蚴感染小鼠模型。3个月后处死小鼠,采集肝脏标本,提取肝脏总RNA,采用RT-qPCR法检测miR-100、miR-125b、miR-126和miR-122的相对表达量,预测并初步分析差异表达miRNA的靶基因。结果实验组小鼠在感染3月肝内miR-122相对表达量为6.335±1.815,是对照组的2.41倍(P<0.05);miR-100、miR-125b和miR-126相对表达量实验组与对照组比较差异无统计学意义(P>0.05)。预测hfe是miR-122的直接靶基因,参与肝脏铁离子代谢的调节。结论小鼠泡球蚴感染中期肝脏miR-122表达上调,推测其通过下调hfe等基因的表达影响肝脏铁离子和脂类代谢,造成肝损伤。
Objective To observe the expression of miR-100, miR-125b, miR-126 and miR-122 in the liver of metaphase (March) mouse with metacercariae infection and to explore its significance. Methods The liver of mice was punctured with the naked eye under direct vision and injected with homogenate of 0.1 ml Echinococcus granulosus to establish a mouse model of Cysticercus by infection. After 3 months, the mice were sacrificed and the liver samples were collected for total RNA extraction. The relative expression levels of miR-100, miR-125b, miR-126 and miR-122 were detected by RT-qPCR and differentially expressed miRNA Of target genes. Results The relative expression of miR-122 in experimental group was 6.335 ± 1.815, which was 2.41 times of the control group (P <0.05). The relative expression of miR-100, miR-125b and miR-126 in experimental group Compared with the control group, there was no significant difference (P> 0.05). It is predicted that hfe, a direct target gene of miR-122, is involved in the regulation of hepatic iron metabolism. Conclusions Up-regulation of miR-122 expression in metaphase liver cells of metacercariae infected mice is presumed to affect hepatic iron and lipid metabolism by down-regulating the expression of hfe and other genes, resulting in hepatic injury.
