目的:观察首乌、洁脉冲剂及其组分对平滑肌细胞体外药敏效应。方法:从“复方-单味药-活性成分”对比分析入手,采用MTT法测定首乌、洁脉冲剂、大黄素及大黄酸对平滑肌细胞增殖抑制作用。结果:首乌、洁脉冲剂与大黄素的有效剂量范围为其血药浓度的25~125倍;大黄酸有效剂量范围为其血药浓度的12.5~75倍。洁脉冲剂、大黄素对平滑肌细胞增殖抑制率分别为21.54%~96.89%,24.6%~94.58%;而首乌、大黄酸效果较差,抑制率分别为16.56%~86.57%,14.38%~85.64%。并且随作用时间延长、药物浓度的增加抑制率都有一定的提高。结论:本研究为首乌、洁泳冲剂、大黄素及大黄酸的医学应用提供了理论依据,为中药复方药效学研究提供了一种可借鉴的方法。
OBJECTIVE: To observe the in vitro drug sensitivity of Shouwu and Jieji pulse agents and their components on smooth muscle cells. Methods: Starting from the comparative analysis of “compound-single-drug-active ingredient”, MTT method was used to determine the inhibitory effect of Shouwu, Jiezheng pulse, emodin and rhein on the proliferation of smooth muscle cells. RESULTS: The effective dosage range of Shouwu and Jiezheng pulse agents and emodin was 25 to 125 times of its blood drug concentration, and the effective dosage range of rhein was 12.5 to 75 times of its blood drug concentration. The inhibitory rates of Jiezheng pulse and emodin on smooth muscle cell proliferation were 21.54%-96.89% and 24.6%-94.58%, respectively. The effects of Shouwu and Rhubarb were poor and the inhibition rates were 16. 56% to 86.57%, 14.38% to 85.64%. And with the extension of the action time, the inhibition rate of the increase of drug concentration has a certain increase. Conclusion: This study provides a theoretical basis for the medical application of Shouwu, Jieying Granules, emodin and rhein, and provides a reference for the pharmacodynamics study of traditional Chinese medicine compound.
