目的:说明丹参酮ⅡA对血管合成血管紧张素Ⅱ及醛固酮的作用。方法:用大鼠肠系膜动脉离体灌注、高效液相色谱法和放射免疫法检测肠系膜动脉中合成的血管紧张素Ⅱ及醛固酮。用逆转录聚合酶链反应(RT-PCR)及SouthernBlot法检测血管中醛固酮合酶基因CYP11B2信使核糖核酸(mRNA)的表达情况。结果:丹参酮ⅡA抑制血管合成醛固酮,但不能抑制血管合成血管紧张素Ⅱ。丹参酮ⅡA抑制血管中醛固酮合酶基因CYP11B2mRNA的表达。结论:丹参酮ⅡA治疗心血管疾病的机制可能与其抑制血管合成醛固酮有关。
Objective: To elucidate the effect of tanshinone IIA on angiogenesis of angiotensin II and aldosterone. METHODS: Angiotensin II and aldosterone synthesis in mesenteric arteries were detected by in vitro perfusion of rat mesenteric artery, high performance liquid chromatography, and radioimmunoassay. The mRNA and protein expression of aldosterone synthase gene CYP11B2 in blood vessels was detected by reverse transcription-polymerase chain reaction (RT-PCR) and Southern Blot. RESULTS: Tanshinone IIA inhibited the synthesis of aldosterone in blood vessels but did not inhibit the synthesis of angiotensin II by blood vessels. Tanshinone II A inhibits the expression of aldosterone synthase gene CYP11B2 mRNA in blood vessels. CONCLUSION: The mechanism of tanshinone II A in the treatment of cardiovascular disease may be related to its inhibition of vascular aldosterone synthesis.
