目的探讨~(99m)Tc-转铁蛋白(Tf)对肝细胞癌的诊断价值。方法以~(99m)Tc标记Tf进行Balb/c小鼠体内生物分布,对Tf受体阻断前后的Balb/c人肝癌细胞系SMMC-7721nu/nu裸鼠进行显像研究,测定SMMC-7721细胞膜Tf受体数及125I-Tf与Tf受体的解离常数(Kd)。结果~(99m)Tc-Tf主要分布于肝、血及肾脏。荷人肝癌SMMC-7721裸鼠注入~(99m)Tc-Tf后18h,肿瘤/血、肿瘤/肝及肿瘤/肌肉的比值分别为24.2、1.7和35.5,非标记Tf抑制前后小鼠肿瘤摄取率的比值为16.3。人肝癌SMMC-7721细胞膜Tf受体数为2.4×105/细胞,Kd为4.46nmol/L。结论~(99m)Tc-Tf在高表达Tf受体的肿瘤病变部位特异性聚积,~(99m)Tc-Tf有望用于肝细胞癌Tf受体显像。
Objective To investigate the diagnostic value of ~ (99m) Tc-transferrin (Tf) in hepatocellular carcinoma. Methods Balb / c mice were biodistributed with ~ (99m) Tc labeled Tf and the Balb / c human hepatocellular carcinoma cell line SMMC-7721nu / nu before and after Tf receptor blockade was studied. SMMC-7721 Cell membrane Tf receptor number and 125I-Tf Tf receptor dissociation constant (Kd). Results ~ (99m) Tc-Tf mainly distributed in liver, blood and kidney. Tumor / blood, tumor / liver and tumor / muscle ratios were 24.2, 1.7 and 35.5 at 18 h after injection of ~ (99m) Tc-Tf in nude mice bearing human hepatoma SMMC-7721, respectively. The ratio is 16.3. The number of Tf receptors in human hepatocellular carcinoma SMMC-7721 cell membrane was 2.4 × 105 / cell, Kd was 4.46nmol / L. Conclusion The ~ (99m) Tc-Tf can accumulate specifically in the tumor site with high Tf receptor expression, and ~ (99m) Tc-Tf is expected to be used for Tf receptor imaging in hepatocellular carcinoma.
