目的:建立中药决明子的高效液相指纹图谱,比较决明子药材、粉体和粉粒指纹图谱的差异。方法:采用Phenomenex C18(4.6 mm×250 mm,5μm)色谱柱,柱温30℃,以乙腈-0.1%磷酸水溶液为流动相梯度洗脱,检测波长280 nm,流速0.8 mL/min。采用中药指纹图谱分析系统/数据库评价软件,对12批不同批次的决明子药材、超微粉体和破壁粉粒指纹图谱进行相似度计算,并对三者的相似度进行比较分析。结果:各批次决明子的指纹图谱基本一致,相似度均>90%,鉴定出27个共有峰,各峰分离度良好,共有峰的相对保留时间的RSD均<2.0%。粉体与药材的指纹图谱基本相似,粉粒与药材的指纹图谱差异较大。结论:本方法具有良好的精密度、重复性、稳定性,各共有峰之间分离度高,可用于决明子药材的真伪鉴别和质量综合评价,为中药决明子的临床应用提供理论参考。
OBJECTIVE: To establish a high performance liquid chromatography (HPLC) fingerprint of Chinese traditional medicine Cassia to compare the difference of the fingerprint of Cassia seed, powder and powder. Methods: The chromatographic column was Phenomenex C18 (4.6 mm × 250 mm, 5 μm). The column temperature was 30 ℃ and acetonitrile-0.1% phosphoric acid was used as mobile phase. The detection wavelength was 280 nm and the flow rate was 0.8 mL / min. The fingerprints of traditional Chinese medicine fingerprinting analysis system / database evaluation software were used to calculate the similarity between 12 batches of different varieties of cassia seed, ultrafine powder and broken powder fingerprints, and the similarity of the three were compared. Results: The fingerprints of cassia seed were consistent in all fingerprints, the similarity was> 90%, 27 common peaks were identified, and the peak separation was good. The relative retention time of common peaks were all lower than 2.0%. Fingerprints of powder and medicinal materials are similar, and the fingerprints of powder and medicinal materials are quite different. Conclusion: The method has good precision, repeatability and stability, high separation between the common peaks, can be used for the authenticity identification and quality evaluation of cassia seed, and provide a theoretical reference for the clinical application of Chinese traditional medicine cassia seed.
