异丙酚复合川芎嗪对大鼠肝脏缺血/再灌注损伤的作用

目的探讨异丙酚复合川芎对大鼠肝脏缺血/再灌注损伤的作用及其机制。方法50只雄性SD大鼠随机分5组,每组10只,即Sham、Mod、Pro、Lig、Pro+Lig组。各组均在阻断第一肝门前20min用药,Pro组经尾静脉持续输注异丙酚20mg·kg-1·h-1,Lig组经尾静脉注射川芎嗪60mg·kg-1,Pro+Lig组经尾静脉注射川芎嗪60mg·kg-1后,经尾静脉持续输注异丙酚20mg·kg-1·h-1;Sham、Mod组分别经尾静脉持续输注等容量生理盐水。在阻断第一肝门前即刻停止输注药物。分别在肝门阻断30min、再灌注60min取腹主动脉血,测定血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、乳酸脱氢酶(LDH)活性。取血后立即处死大鼠,测定肝组织丙二醛(MDA)含量、超氧化物歧化酶(SOD)、黄嘌呤氧化酶(XOD)活性,并观察肝细胞超微结构。结果Mod、Pro、Lig、Pro+Lig组血清ALT、AST活性均高于Sham组(P<0.01),Pro、Lig、Pro+Lig组血清ALT、AST活性均低于Mod组(P<0.05或0.01),Pro+Lig组血清ALT。活性均低于Pro、Lig组(P<0.05)。与Sham组比较,Mod、Pro、Lig组肝组织MDA含量升高,Mod组肝组织SOD活性降低,Pro、Lig、Pro+Lig组肝组织SOD活性升高,Mod、Pro、Lig、Pro+Lig组肝组织XOD活性升高(P<0.05或0.01);与Mod组比较,Pro、Lig、Pro+Lig组肝组织MDA含量降低,SOD活性升高,Pro+L Objective To investigate the effect and mechanism of propofol and ligusticum chuanxiong on liver ischemia / reperfusion injury in rats. Methods Fifty male Sprague-Dawley rats were randomly divided into 5 groups (n = 10), Sham, Mod, Pro, Lig and Pro + Lig groups. In each group, propofol 20 mg · kg-1 · h-1 was infused continuously through caudal vein in the Pro group, ligustrazine 60 mg · kg-1 + Lig group were injected intravenously with ligustrazine 60mg · kg-1 through the tail vein, and propofol 20mg · kg-1 · h-1 was infused through the tail vein continuously. Sham and Mod rats were continuously infused with equal volumes of normal saline . Infusions were stopped immediately before the first hepatic portal was blocked. Abdominal aorta blood was collected for 30 minutes after hepatic portal vein blockade and 60 minutes after reperfusion, respectively. Serum alanine aminotransferase (ALT), aspartate aminotransferase (AST) and lactate dehydrogenase (LDH) activity were measured. The rats were sacrificed immediately after the blood was taken and the content of malondialdehyde (MDA), superoxide dismutase (SOD) and xanthine oxidase (XOD) in the liver tissues were measured. The ultrastructure of hepatocytes was observed. Results The serum levels of ALT and AST in Mod, Pro, Lig and Pro + Lig groups were significantly higher than those in Sham group (P <0.01 or P <0.01), while the levels of ALT and AST in Pro, Lig and Pro + Lig groups were lower than those in Mod group 0.01), serum ALT in Pro + Lig group. Activity were lower than Pro, Lig group (P <0.05). Compared with Sham group, MDA content in liver increased in Mod, Pro and Lig groups, SOD activity in liver decreased in Mod group, and increased in Pro, Lig and Pro + Lig groups. Mod, Pro, Lig, Pro + Lig Compared with Mod group, the content of MDA in liver tissue of Pro, Lig, Pro + Lig group decreased and the activity of SOD increased, while that of Pro + L group increased (P <0.05 or 0.01)
