目的 通过观察APP17肽和谷氨酸对人神经母细胞瘤株SY5Y生长及NT 3、NF表达的影响 ,初步证实APP17肽对谷氨酸引起的神经元毒性具有一定保护效应。方法 MTT代谢率、LDH漏出率测定、细胞计数 ,WesternBlot观察SY5YNT 3、NF表达。结果 谷氨酸可使MTT代谢率降低、细胞计数减少 ,LDH漏出率升高 ,NT 3、NF表达减少 ,加入APP17肽可使上述指标恢复或接近正常。结论 谷氨酸对神经元有毒性作用 ,而APP17肽可减轻这种损伤 ,对神经元产生保护作用。
Objective To observe the effect of APP17 peptide and glutamate on the growth of human neuroblastoma SY5Y and the expression of NT 3 and NF, to initially confirm that APP17 peptide has a protective effect on glutamate-induced neuronal toxicity. Methods MTT metabolic rate, LDH leakage rate, cell count, Western-blot were used to observe the expression of SY5YNT 3 and NF. Results Glutamate could decrease the metabolic rate of MTT, reduce the cell count, increase the leakage rate of LDH, reduce the expression of NT 3 and NF. The addition of APP17 peptide can restore or close the normal expression of these indexes. Conclusion Glutamate has a toxic effect on neurons, whereas APP17 peptide can reduce this damage and protect neurons.
