目的检测人类X射线交错互补修复基因(X-ray repair cross-compplementing gene1,XRCC1)、人类8-羟基鸟嘌呤糖苷酶1(human 8-hydroxyguanine glycosylase 1,hOGG1)基因和人类X射线交错互补修复基因(X-rayrepair cross-compplementing gene 3,XRCC3)蛋白在食管癌和癌旁组织中的表达,探讨3种蛋白与食管癌发生及发展的关系。方法采用蛋白免疫印迹法检测XRCC1、XRCC3及hOGG1蛋白在食管癌与癌旁组织中的表达,应用ECL法显影并以其灰度值来表示蛋白表达的量。结果 XRCC1、XRCC3及hOGG1蛋白在癌组织中的表达高于癌旁组织(P均<0.05)。同时XRCC1蛋白在癌组织与癌旁组织中的差异程度与民族有相关性(P<0.05)。结论 XRCC1、XRCC3及hOGG1蛋白表达水平与食管癌的发生可能具有相关性,并且XRCC1蛋白还可能参与食管癌的进展。
Objective To detect human X-ray repair cross-complementing gene1 (XRCC1), human 8-hydroxyguanine glycosylase 1 (hOGG1) gene and human X-ray intercalator repair gene (XRCC3) protein in esophageal and paracancerous tissues, and to explore the relationship between the three proteins and the occurrence and development of esophageal cancer. Methods Western blotting was used to detect the expression of XRCC1, XRCC3 and hOGG1 protein in esophageal carcinoma and paracancerous tissues. The expression of XRCC1, XRCC3 and their adjacent tissues was evaluated by ECL method. Results The expression of XRCC1, XRCC3 and hOGG1 protein in cancer tissues was higher than that in adjacent tissues (all P <0.05). At the same time, the difference of XRCC1 protein in cancer tissues and adjacent non-cancerous tissues was related to ethnicity (P <0.05). Conclusion The expression of XRCC1, XRCC3 and hOGG1 may be correlated with the occurrence of esophageal cancer, and the expression of XRCC1 may be involved in the progression of esophageal cancer.
