目的:建立HPLC法测定制痂酊中儿茶素和表儿茶素含量的方法,同时考察不同剂量~(60)Co辐照对其含量影响。方法:色谱柱:Kromasil C_(18)(250 mm×4.6 mm,5μm),流动相:2.2%冰醋酸-乙腈(86:14),流速:0.8 ml·min~(-1),检测波长:280nm,柱温:35℃。结果:儿茶素和表儿茶素分别在1.025~12.813μg,0.196~2.450μg范围内线性良好,加样回收率分别为98.0%,99.8%(n=5);~(60)Co辐照剂量为3 kGy时,儿茶素和表儿茶素含量分别减少2.77%,3.43%,随着辐照剂量的增加,两者含量有逐渐下降的趋势。结论:对单批次样本测定表明:~(60)Co辐照对制痂酊中儿茶素和表儿茶素的含量有一定影响,但在低剂量(≤3 kGy)时影响不显著。
OBJECTIVE: To establish a HPLC method for the determination of catechin and epicatechin in scab tincture, and to study the effects of different doses of ~ (60) Co irradiation on its content. METHODS: Kromasil C18 (250 mm × 4.6 mm, 5 μm) was used as the mobile phase. The mobile phase consisted of 2.2% glacial acetic acid and acetonitrile (86:14), the flow rate was 0.8 ml · min ~ 280 nm, column temperature: 35 ° C. Results: The linearities of catechin and epicatechin were 1.025 ~ 12.813μg and 0.196 ~ 2.450μg, respectively. The recoveries were 98.0% and 99.8% respectively (n = 5) At the dose of 3 kGy, the contents of catechins and epicatechin decreased by 2.77% and 3.43%, respectively. With the increase of irradiation dose, the content of both catechins and epicatechin decreased gradually. CONCLUSION: The results of single batch test showed that the irradiation of ~ (60) Co has a certain effect on the content of catechin and epicatechin in the scab tincture, but it has no significant effect at low dose (≤3 kGy).
