建立以槲皮素为标准品用荧光分光光度法测定荷叶中总黄酮含量的新方法,荷叶黄酮用超声波提取。试验结果表明:当槲皮素的浓度范围在6.7×10-6~6×10-4g/L之间时,荧光强度和槲皮素浓度呈良好的线性关系,建立的线性回归方程为y=531.84048x-3.54448,R2=0.99903,该方法的检出限为2.51×10-7g/L;荷叶中总黄酮的平均含量为0.2452mg/g,RSD=1.696%。该方法操作简便,准确度高,可以用于荷叶的质量控制。
A new method for the determination of total flavonoids in lotus leaf with quercetin as standard by fluorescence spectrophotometry was established. The flavonoids in lotus leaf were extracted by ultrasonic wave. The results showed that when the concentration range of quercetin was between 6.7 × 10-6 ~ 6 × 10-4g / L, the linear relationship between the fluorescence intensity and the concentration of quercetin showed a good linearity. The linear regression equation established was y = The detection limit of this method was 2.51 × 10-7g / L. The average content of total flavonoids in lotus leaf was 0.2452mg / g, RSD = 1.696%. The method is simple, accurate and can be used for the quality control of lotus leaf.
