miR-155对子痫前期滋养层细胞炎症应答的调控研究

目的观察子痫前期胎盘组织中miR-155和炎症因子的表达情况,探讨该miR对子痫前期炎症应答的调控。方法收集2014年1月至2015年12月在深圳市罗湖区人民医院住院的子痫前期患者和同期住院正常晚期妊娠孕妇各20例,应用Real-time PCR检测胎盘组织中miR-155和白细胞介素-1β、白细胞介素-6、肿瘤坏死因子-α的表达水平;分离培养胎盘滋养层细胞,采用miR-155抑制剂(anti-miR-155)敲低miR-155表达,通过real-time PCR检测IL-1β、IL-6、TNF-α的表达变化情况。结果子痫前期组胎盘组织内miR-155和IL-1β、IL-6、TNF-α表达量较正常对照组明显升高,敲低miR-155后显著抑制IL-1β、IL-6、TNF-α的表达。结论 miR-155参与调控子痫前期滋养层细胞IL-1β、IL-6、TNF-α的表达,从而加剧子痫前期炎症反应。 Objective To observe the expression of miR-155 and inflammatory cytokines in placenta of preeclampsia and to explore the regulation of the miR on the inflammatory response in preeclampsia. Methods Twenty patients with preeclampsia and 20 normal pregnant women in the same period were enrolled from January 2014 to December 2015 in Shenzhen People’s Hospital of Luohu District. Real-time PCR was used to detect the expression of miR-155 and interleukin 1β, IL-6 and TNF-α. The placental trophoblast cells were isolated and cultured. The expression of miR-155 was knocked down by using miR-155 inhibitor (anti-miR-155) PCR detection of IL-1β, IL-6, TNF-α expression changes. Results The levels of miR-155, IL-1β, IL-6 and TNF-α in placenta of preeclampsia group were significantly higher than those in normal control group. Knockdown of miR-155 significantly inhibited the expression of IL-1β, IL- -α expression. Conclusion miR-155 is involved in the regulation of the expression of IL-1β, IL-6 and TNF-α in trophoblast cells in preeclampsia, which may exacerbate the inflammatory response in preeclampsia.
