从羊肚菌发酵的菌丝体中提取粗多糖,经DEAE-Sepharose F.F和Superdex 200柱层析纯化得到MEP-Ⅱ,琼脂糖电泳法和HPLC法鉴定其为均一组分。凝胶过滤法测得其分子质量为2.8×10~4u,比色法测其总糖含量、蛋白含量、糖醛酸含量分别为87.5%、8.4%和23.2%。红外光谱呈现多糖的特征吸收峰,含有β-吡喃糖苷键。气相色谱法得其单糖组分为鼠李糖(Rha)、半乳糖(Gal)、葡萄糖(Glc)、甘露糖(Man),它们的摩尔比为2.1:11.3:35.6:1.0,β-消去反应初步表明其存在O-糖肽键。
The crude polysaccharide was extracted from mycelium fermented by Morchella esculenta and purified by column chromatography on DEAE-Sepharose F.F and Superdex 200 to obtain MEP-II. The agarose electrophoresis method and HPLC method were used to identify the homogenate. The gel filtration method had a molecular mass of 2.8×10 4 u. The total sugar content, protein content, and uronic acid content were 87.5%, 8.4%, and 23.2%, respectively. The infrared spectrum shows the characteristic absorption peak of the polysaccharide, containing the β-pyranoside bond. The monosaccharide components of the gas chromatography were Rham, Gal, Glc, and Man, with a molar ratio of 2.1:11.3:35.6:1.0 and β-elimination. The reaction initially showed its presence of an O-glycopeptide bond.
