目的 扩增霍乱弧菌不同血清型I类整合子基因片段 ,并分析I类整合子在霍乱弧菌O1和O13 9血清型中的分布。 方法 收集霍乱弧菌O1和O13 9血清型 40株 ,根据GenBank资料设计PCR引物 ,扩增霍乱弧菌I类整合子基因片段 ,并用限制性酶切分析法进行鉴定 ,分析I类整合子在不同血清型的分布情况。 结果 从霍乱弧菌O1和O13 9血清型中均能扩增约 80 0bp的基因片段 ,扩增片段经PstI酶切后可获得 671和 12 7bp的片段 ,经HindIII酶切后可获得 5 80、160和 5 8bp的片段 ,经HindIII和PstI酶切后可获得 45 3、160、12 7和 5 8bp的片段 ,I类整合子在O13 9血清型霍乱弧菌中分布多于O1血清型。 结论 I类整合子与霍乱散发菌株耐药性的传递、血清型的分布有密切的联系。
Objective To amplify the I serotype I integron gene of Vibrio cholerae and to analyze the distribution of class I integrons in Vibrio cholerae O1 and O139 serotypes. Methods 40 strains of Vibrio cholerae O1 and O139 serotypes were collected. PCR primers were designed according to GenBank data to amplify the gene fragment of Vibrio cholerae class I integron and identified by restriction analysis. Serum distribution. Results A total of 80 0bp fragment was amplified from Vibrio cholerae O1 and O139 serotypes. The amplified fragments were 671 and 12 7 bp after digestion with PstI. After digestion with HindIII, 160 and 58 bp fragments were digested with HindIII and PstI to obtain 45 3,160,127 and 5 8bp fragments. The class I integrins distributed more in the O139 serotype Vibrio cholerae than in the O1 serotypes. Conclusions Class I integrons are closely linked to the transmission of drug resistance and the distribution of serotypes in cholera-infected strains.
