目的:探讨利心冲剂对心衰大鼠模型抗凋亡作用机制。方法:腹腔注射阿霉素(ADR)复制大鼠心衰模型,左心室插管术测定血流动力学指标,Real time PCR检测凋亡相关基因。结果:利心冲剂治疗组和模型组比较,左心室收缩内压显著升高;凋亡相关基因Bcl-2、Bcl-2/bax增高;均有显著性差异(P<0.05),利心冲剂治疗组心肌细胞变性坏死明显减轻。结论:利心冲剂提高凋亡相关基因Bcl-2、Bcl-2/bax表达,减轻心肌细胞凋亡,改善心脏收缩与舒张功能。
Objective: To explore the mechanism of anti-apoptosis effect of Lixin granule on rat heart failure model. METHODS: Rat heart failure model was induced by intraperitoneal injection of adriamycin (ADR), hemodynamic parameters were measured by left ventricular catheterization, and apoptosis-related genes were detected by Real time PCR. Results: The systolic pressure of the left ventricle was significantly higher in the Lixin granule treatment group than in the model group; the apoptosis-related genes Bcl-2 and Bcl-2/bax were increased; there were significant differences (P<0.05). Degeneration and necrosis of cardiomyocytes in the treatment group were significantly reduced. Conclusion: Lixin Granule can increase the expression of apoptosis-related genes Bcl-2, Bcl-2/bax, reduce myocardial cell apoptosis, and improve cardiac contractility and diastolic function.
