目的:探讨在脓毒症模型中雷公藤内酯醇的抗炎及免疫调节的双重作用效应。方法:60只SD大鼠以盲肠结扎手术制作脓毒症模型,分为假手术组、模型组及雷公藤内酯醇组。采用ELISA检测各组血IL-6、IL-1及TNF-α水平,RT-PCR定量检测各组肺组织中TLR4mRNA及NF-κBp65mRNA的表达,比较各组中肺的湿/干比值。流式细胞仪检测CD4~+/CD8~+比值变化。结果:与假手术组比,模型组中IL-6、IL-1及TNF-a含量增高,肺组织中TLR-4mRNA、NF-κBp65mRNA的表达均增高,CD4~+/CD8~+的含量降低,湿/干肺比值升高,各组差异有统计学意义(P<0.05)。与模型组相比,雷公藤内酯醇组中IL-1、IL-6及TNF-α水平.肺组织中TLR4mRNA及NF-κBp65mRNA的表达,湿/干肺比值均明显降低,CD4~+/CD8~+的含量增高,差异有统计学意义(P<0.05)。结论:在脓毒症中雷公藤内酯醇能抑制炎症级联反应和调节T淋巴细胞亚群CD4~+/CD8~+比值发挥抗炎及调节免疫平衡状态的治疗作用。
Objective: To investigate the anti-inflammatory and immunoregulatory effects of triptolide in sepsis model. Methods: Sixty Sprague-Dawley rats were operated on cecal ligation to make sepsis model and divided into sham operation group, model group and triptolide group. The levels of IL-6, IL-1 and TNF-α in blood were measured by ELISA. The expressions of TLR4 mRNA and NF-κBp65 mRNA were detected by RT-PCR. The wet / dry ratio of lung in each group was compared. Flow cytometry detection of CD4 ~ + / CD8 ~ + ratio changes. Results: The levels of IL-6, IL-1 and TNF-α in the model group were significantly higher than those in the sham operation group. The expressions of TLR-4 mRNA and NF-κBp65 mRNA in the lung tissue were increased and the contents of CD4 + / CD8 + , Wet / dry lung ratio increased, the differences were statistically significant (P <0.05). The levels of IL-1, IL-6 and TNF-α in the triptolide-treated group were significantly lower than those in the untreated group, and the expressions of TLR4mRNA and NF-κBp65mRNA, wet / ~ + Content increased, the difference was statistically significant (P <0.05). Conclusion: Triptolide can inhibit the inflammatory cascade and regulate the ratio of CD4 ~ + / CD8 ~ + in T lymphocyte subsets in sepsis to exert its anti-inflammatory and immunomodulatory effects.
