探讨用植物血凝素(PHA)诱导人外周血单核的细胞(PBMC)产生白细胞介素-2(IL-2)的实验条件,并用PHA诱导正常人PBMC产生IL-2及用IL-2依赖细胞株(CTLL-2)增殖试验来滴定IL-2活性单位。结果:(1)PHA-M诱导人PBMO产生IL-2的最适条件:PBMC为1×10~3/ml、PHA-M浓度为0.5~1%、诱生时间为24h、RPMI1640营养液中需加入2~10%灭活的人AB型血清;(2)无论是PHA-M或PHA在较高剂量时(分别≥0.25%和6.25μg/m1)均抑制aTLL-2的生长;(3)用PHA-M诱导19例正常人PBMG产生IL-2,其活性单位为64±65u/mL(±SD,范围4~260)。
To investigate the production of interleukin-2 (IL-2) by human hemagglutinin (PHA) -induced peripheral blood mononuclear cells (PBMCs) and to induce the production of IL-2 by normal human PBMC with IL-2 Dependent cell line (CTLL-2) proliferation assay was used to titrate IL-2 active units. Results: (1) The optimal conditions for PHA-M to induce IL-2 production in human PBMO were: 1 × 10-3 / ml PBMC, 0.5-1% PHA-M and 24 h induction time. 2 to 10% inactivated human type AB serum should be added; (2) either PHA-M or PHA inhibits the growth of aTLL-2 at higher doses (≥ 0.25% and 6.25 μg / ml, respectively); Nineteen normal PBMGs were induced to produce IL-2 by PHA-M with an activity unit of 64 ± 65 u / mL (± SD, range 4-260).
