目的研究人体不同类型甲状腺肿瘤组织中基膜胶原水平的变化规律及其与恶性肿瘤的关系。方法甲状腺肿瘤手术切除术标本(癌旁正常甲状腺组织、甲状腺乳头状癌组织、甲状腺腺瘤组织、结节性甲状腺肿组织)做组织羟脯氨酸(HYP)含量测定、Ⅳ型胶原(ColⅣ)及基质金属蛋白酶-9(MMP-9)蛋白免疫组化染色。结果与癌旁正常甲状腺组织比较,甲状腺乳头状癌组织中HYP含量显著升高(P<0.01),其余两种组织中HYP含量没有显著改变(P>0.05);甲状腺肿瘤组织中ColⅣ蛋白表达显著下调而MMP-9蛋白表达呈显著增多的趋势(P<0.01或P<0.05);且甲状腺乳头状癌组织中MMP-9蛋白水平显著高于其余2组(P<0.05)。结论甲状腺肿瘤组织中,基膜成分的减少反馈性启动了细胞内胶原蛋白的大量合成。结果提示,组织内HYP含量升高及基膜成分的减少与恶性甲状腺肿瘤相关。
Objective To study the changes of collagen level in basement membrane of different types of human thyroid tumors and its relationship with malignant tumors. Methods Tissue Hydroxyproline (HYP) content was measured in thyroid tumor surgically resected specimens (adjacent normal thyroid tissue, thyroid papillary carcinoma, thyroid adenoma and nodular goiter) And matrix metalloproteinase-9 (MMP-9) protein immunohistochemical staining. Results Compared with the adjacent normal thyroid tissue, the content of HYP in papillary thyroid carcinoma was significantly increased (P <0.01), and the content of HYP in the other two tissues did not change significantly (P> 0.05). In thyroid tumor (P <0.01 or P <0.05). The expression of MMP-9 protein in papillary thyroid carcinoma was significantly higher than that in the other two groups (P <0.05). Conclusions In thyroid tumors, the reduced feedback of basement membrane components initiates the massive synthesis of intracellular collagen. The results suggest that elevated tissue HYP content and basement membrane composition and the reduction of malignant thyroid tumors.
