对 1 9例慢性支气管炎 (慢支 )缓解期患者和1 5例正常对照者经局部支气管肺泡灌洗获取的肺泡巨噬细胞 ( AM) ,加川芎嗪 ( ligustrazine) ,脂多糖( LPS)体外培养 2 4 h后 ,以流式细胞仪分析其CD1 1 c和 CD1 4表达的百分率 ;测定其胞浆游离钙水平 .慢支组 AM膜上 CD1 1 c和 CD1 4的表达均显著高于正常对照组 ;1 0 0 mg· L-1LPS可使慢支组AM膜上 CD1 1 c的表达进一步增加 ;40 0 mg· L-1川芎嗪可明显减少 LPS诱导慢支组 AM膜上CD1 1 c的过度表达 .慢支组 AM胞浆中基础钙水平较正常对照组明显增高 ,但其静息钙水平与正常对照组无显著差异性 ;40 0 mg· L-1川芎嗪明显抑制1 0 mg· L-1LPS诱导慢支 AM胞浆游离钙水平的升高 .结果提示川芎嗪通过抑制慢支患者 AM胞浆游离钙水平升高下调其 CD1 1 c的表达 ,可能对慢支缓解期气道内非特异性炎症有抑制作用
The bronchial alveolar macrophages (AM), ligustrazine and lipopolysaccharide (LPS) obtained from local bronchoalveolar lavage in 19 patients with chronic bronchitis (chronic bronchitis) and 15 normal controls were studied in vitro After culturing for 24 h, the percentage of CD1 1 c and CD1 4 expression was analyzed by flow cytometry, and the level of cytoplasmic free calcium was measured. The expressions of CD1 1 c and CD14 on AM membrane in chronic bronchitis group were significantly higher than those in normal The control group; the expression of CD11c on AM membrane of chronic bronchitis group was further increased by 100 mg · L-1 LPS; Ligustrazine at the concentration of 40 0 mg · L-1 could obviously reduce the expression of CD1 1 c .The level of basal calcium in AM cytoplasm in chronic bronchitis group was significantly higher than that in normal control group, but the resting calcium level was not significantly different from that in normal control group. Tetramethylpyrazine 40 0 mg · L-1 significantly inhibited 10 mg L-1 LPS induced elevation of free cytoplasm calcium in chronic bronchitis patients.Results: Ligustrazine could decrease the expression of CD11c in AM cytoplasm by inhibiting the increase of intracellular free calcium in chronic bronchitis patients, Non-specific inflammation is inhibited
