目的:探讨干扰素(IFN)对肝癌细胞HLA-DR表达的调节作用。方法:采用免疫组化ABC法和ELISA法分别检测了不同剂量的IFN-γ或IFN-α刺激前后HLA-DR在HHCC、SMMC-7721、BEL-7402和HCC-9204等4种人肝细胞癌细胞系以及正常人肝细胞系QZG的表达情况。结果:IFN-γ或IFN-α刺激之前,5种细胞系中只有少数的SMMC-7721细胞可表达HLA-DR;刺激之后,对HLA-DR的表达5种细胞系都有所增强,并且与IFN-γ的剂量呈正相关,QZG不如肝癌细胞明显,IFN-γ的作用强于IFN-α。结论:IFN可以上调肝癌细胞对HLA-DR的表达。关键词:##4干扰素;;HLA-DR;;肝细胞癌;;免疫组织化学;;ELISA
Objective: To investigate the effect of interferon (IFN) on the regulation of HLA-DR expression in hepatoma cells. METHODS: Immunohistochemical ABC method and ELISA method were used to detect four human hepatocellular carcinomas of HLA-DR before and after stimulation with different doses of IFN-γ or IFN-α in HHCC, SMMC-7721, BEL-7402 and HCC-9204, respectively. Expression of QZG in cell lines and normal human hepatic cell lines. Results: Before stimulation with IFN-γ or IFN-α, only a small number of SMMC-7721 cells in 5 cell lines could express HLA-DR; after stimulation, the 5 cell lines of HLA-DR expression were enhanced, and The dose of IFN-γ was positively correlated, QZG was not as obvious as hepatoma cells, and IFN-γ was stronger than IFN-α. Conclusion: IFN can upregulate the expression of HLA-DR in liver cancer cells. Key words: ##4 interferon; HLA-DR; hepatocellular carcinoma; immunohistochemistry; ELISA
