目的观察槲皮素对胃癌细胞SGC-7901生长及HSP70和EGFR的影响。方法以台盼蓝拒染法计数胃癌细胞的生长抑制率,透射电镜从形态变化上了解凋亡的发生,流式细胞术检测细胞周期变化,免疫组化检测HSP70和EGFR的表达。结果台盼蓝拒染法计数显示槲皮素抑制SGC-7901细胞增殖的作用明显,且呈浓度和时间依赖性,槲皮素处理48 h后的IC50为14.12μmol/L。形态学检测显示出细胞凋亡的特征变化,流式细胞仪检测表明经10-20μmol/L的槲皮素处理,SGC-7901细胞周期阻滞于G0/G1期,经10μmol/L槲皮素处理的SGC-7901下调HSP70和EGFR蛋白的表达。结论槲皮素能抑制胃癌细胞的生长,呈时间剂量依赖性,能诱导SGC-7901发生凋亡,使细胞周期阻滞于G0/G1期,抑制增生与诱导凋亡的机制可能与下调HSP70和EGFR的表达有关。
Objective To observe the effect of quercetin on the growth of gastric cancer cell SGC-7901 and the effects of HSP70 and EGFR. Methods The growth inhibition rate of gastric cancer cells was counted by trypan blue exclusion. The morphological changes of apoptosis were observed by transmission electron microscopy. The changes of cell cycle were detected by flow cytometry. The expressions of HSP70 and EGFR were detected by immunohistochemistry. Results The results of trypan blue exclusion assay showed that quercetin inhibited the proliferation of SGC-7901 cells in a dose-dependent and time-dependent manner. The IC50 of quercetin 48 h after treatment was 14.12 μmol / L. Morphological examination revealed the characteristic changes of apoptosis. Flow cytometry showed that the cell cycle of SGC-7901 cells was arrested in G0 / G1 phase after 10-20μmol / L quercetin treatment. After 10μmol / L quercetin Treated SGC-7901 down-regulated HSP70 and EGFR protein expression. Conclusions Quercetin can inhibit the growth of gastric cancer cells in a dose-and time-dependent manner, induce apoptosis of SGC-7901 cells, block the cell cycle in G0 / G1 phase, inhibit the proliferation and induce apoptosis mechanisms may be related to the downregulation of HSP70 and EGFR expression.
