目的研究丙烯酰胺(ACR)亚慢性染毒对小脑神经丝(NF)蛋白的影响,探讨其中毒性神经病机制。方法以204、0 mg/kg ACR腹腔注射雄性Wistar大鼠2个月,Western Blot法测定小脑中NF-L、NF-M和NF-H相对含量。结果与对照组相比,高剂量组小脑上清液NF-L、NF-M、NF-H和沉淀NF-L分别增高348%、241%、32%和20%(P<0.01),而沉淀NF-H降低14%(P<0.05);低剂量组沉淀NF-L、上清NF-H分别增加16%、17%(P<0.05);低剂量组上清NF-L、NF-M及沉淀NF-M、NF-H均略为降低,但差异无显著性(P>0.05)。结论ACR亚慢性中毒可引起小脑组织中NF亚单位蛋白的明显改变,尤其以上清中NF-L、NF-M升幅最大(348%、241%)。ACR引起的中毒性神经病与小脑组织中NF亚单位的含量变化有关。
Objective To investigate the effect of subchronic ACR on cerebellar neurofilament (NF) protein and to explore the mechanism of toxic neuropathy. Methods Male Wistar rats were injected intraperitoneally with 204,0 mg / kg ACR for 2 months. The relative contents of NF-κB, NF-κB and NF-κ in the cerebellum were measured by Western Blot. Results Compared with the control group, the contents of NF-κL, NF-κB, NF-κL and NF-κ in cerebellum supernatants were increased by 348%, 241%, 32% and 20% (P <0.01) (P <0.05). The contents of NF-L and NF-H in the supernatant increased by 16% and 17% (P <0.05) in the low-dose group, M and NF-M and NF-H were slightly decreased, but the difference was not significant (P> 0.05). Conclusion ACR subchronic poisoning can cause significant changes in NF subunit protein in cerebellum, especially in the supernatant NF-L, NF-M increased the most (348%, 241%). ACR-induced toxic neuropathy is associated with changes in NF subunit content in cerebellum.
