目的探讨ADAM17(a disintegrin and metalloproteinase 17,ADAM17)mRNA在孤立性大肝癌(SLHCC)中的表达及其与病理特征的关系。方法采用逆转录多聚酶链式反应(RT-PCR)半定量检测8例SLHCC、12例小肝癌(SHCC),11例结节性肝癌(NHCC)以及相应癌旁肝组织中ADAM17mRNA的表达。结果NHCC的静脉浸润(83·3%)明显多于SLHCC和SHCC,其分化程度也较SLHCC和SHCC差(P<0·05),而比较SLHCC和SHCC的病理特征未见明显差异。77·4%的肝癌组织中可见ADAM17mRNA表达,明显高于癌旁肝组织(35·5%)。NHCC中ADAM17mRNA的表达高于SLHCC和SHCC(P<0·05)。分化较差的肝癌中ADAM17mRNA表达高于分化较好的肝癌(P<0·05)。结论ADAM17mRNA的表达与肝癌的分化程度密切相关。而SLHCC表达ADAM17mRNA相对较低可能与SLHCC相对良好的分子病理特征有关。
Objective To investigate the expression of ADAM17 (ADAM17) mRNA in isolated large hepatocellular carcinoma (SLHCC) and its relationship with pathological features. Methods Reverse transcription polymerase chain reaction (RT-PCR) was used to detect the expression of ADAM17 mRNA in 8 cases of SLHCC, 12 cases of small hepatocellular carcinoma (SHCC), 11 cases of nodular hepatocellular carcinoma (NHCC) and corresponding paracancerous liver tissues. Results The infiltration rate of NHCC was 83.3% higher than that of SLHCC and SHCC. The differentiation degree of NHCC was also lower than that of SLHCC and SHCC (P <0.05). There was no significant difference between the two groups in the pathological features of SLHCC and SHCC. The expression of ADAM17 mRNA in 77.4% of HCC tissues was significantly higher than that in para-cancerous liver tissues (35.5%). The expression of ADAM17 mRNA in NHCC was higher than that in SLHCC and SHCC (P <0.05). The poorly differentiated HCC ADAM17 mRNA expression was higher than well-differentiated HCC (P <0.05). Conclusion ADAM17 mRNA expression is closely related to the degree of differentiation of HCC. However, the relatively low expression of ADAM17 mRNA in SLHCC may be related to the relatively good molecular pathological features of SLHCC.
