目的研究全氟碳化合物典型代表物质全氟辛酸与全氟辛基磺酸钾28 d重复剂量经口暴露对大鼠的毒理学效应。方法 PFOA与PFOS-K的暴露分别设置4个组别,每组20只SD大鼠,雌雄各半,经口灌胃暴露,每天暴露1次,连续暴露28 d。其中PFOA:溶剂对照(0mg/kg)组、低剂量(5 mg/kg)组、中剂量(20 mg/kg)组及高剂量(100 mg/kg)组;PFOS-K:溶剂对照(0mg/kg)组、低剂量(1 mg/kg)组、中剂量(4 mg/kg)组及高剂量(10 mg/kg)组。暴露结束后取血进行血清生化学检查,取主要脏器计算脏器系数并进行组织病理学检查。结果 PFOA与PFOS-K组动物在暴露过程中体重均明显下降。血清生化检查结果显示PFOS-K对实验动物肝脏与肾脏具有毒性作用。脏器系数计算结果显示PFOA与各剂量组肝脏脏器系数与溶剂组相比显著增加。病理学检查结果显示,PFOA毒性作用的靶器官为肝脏和肺,PFOS-K毒性作用的靶器官为肝脏、肾脏、肺和脾脏。结论 PFOA与PFOS-K在研究剂量下对实验动物具有毒性作用,PFOA毒性作用的靶器官为肝脏和肺,PFOS-K毒性作用的靶器官为肝脏、肾脏、肺和脾脏。
Objective To study the toxicological effects of oral exposure of perfluorooctanoic acid and perfluorooctane sulfonate (KPS), a typical representative of perfluorocarbons, for 28 days in rats. Methods PFOA and PFOS-K were exposed to four groups, 20 SD rats of each group, male and female, exposed to oral gavage once a day for 28 days. PFOA: solvent control (0 mg / kg), low dose (5 mg / kg), medium dose (20 mg / kg) and high dose (100 mg / / kg group, low dose (1 mg / kg) group, medium dose (4 mg / kg) group and high dose (10 mg / kg) group. Serum biochemical tests were performed after the end of exposure to take blood, organ coefficients of major organs were calculated and histopathological examination was performed. Results The body weight of PFOA and PFOS-K animals significantly decreased during exposure. Serum biochemical tests showed that PFOS-K had toxic effects on the liver and kidney of experimental animals. The results of organ coefficient showed that the coefficient of liver organ of PFOA and each dose group increased significantly compared with that of solvent group. The results of pathological examination showed that the target organs of toxic effects of PFOA were liver and lung, and the target organs of toxic effects of PFOS-K were liver, kidney, lung and spleen. Conclusions PFOA and PFOS-K have toxic effects on experimental animals at the doses studied. The target organs of toxic effects of PFOA are liver and lung. The target organs of PFOS-K toxicity are liver, kidney, lung and spleen.
