目的探讨腹腔脂肪组织脂肪细胞因子mRNA表达与新疆维吾尔族肥胖及T2DM的相关性。方法选取腹部择期手术的维吾尔族患者114例,分为正常对照(NC)组50名,肥胖(Ob)组48例和T2DM组16例,术时取网膜脂肪组织,采用RT-PCR方法检测各组APN、TNF-α、MCP-1mRNA表达情况。结果 NC组APN高于Ob、T2DM组,TNF-α低于Ob、T2DM组,MCP-1低于T2DM组(P<0.05)。APN mRNA相对拷贝数与BMI、DBP、FPG、TG、LDL-C水平及TNF-α、MCP-1表达量呈负相关(P<0.05)。TNF-αmRNA相对拷贝数与BMI、FPG、TG呈正相关(P<0.05)。MCP-1mRNA相对拷贝数与体重、BMI、WC呈正相关(P<0.05)。结论腹腔脂肪组织脂肪细胞因子APN、TNF-α、MCP-1mRNA异常表达可能是新疆维吾尔族肥胖及T2DM发生的危险因素。
Objective To investigate the correlation between the expression of adipocytokine mRNA in abdominal adipose tissue and the obesity and T2DM in Uighur in Xinjiang. Methods A total of 114 Uyghur patients undergoing elective abdominal surgery were divided into control group (n = 50), ob group (n = 48) and T2DM group (n = 16). Retinal adipose tissue was obtained during operation and RT- The expression of APN, TNF-αand MCP-1mRNA in each group. Results The APN in NC group was higher than that in Ob and T2DM group, the level of TNF-α was lower than that of Ob and T2DM group, and the level of MCP-1 in NC group was lower than that of T2DM group (P <0.05). The relative copy number of APN mRNA was negatively correlated with the levels of BMI, DBP, FPG, TG, LDL-C and the levels of TNF-α and MCP-1 (P <0.05). The relative copy number of TNF-α mRNA was positively correlated with BMI, FPG and TG (P <0.05). The relative copy number of MCP-1 mRNA was positively correlated with body weight, BMI and WC (P <0.05). Conclusion Abnormal expression of adipocyte cytokines APN, TNF-α and MCP-1 may play an important role in obesity and T2DM in Xinjiang Uygur.
