[目的]探讨转移相关基因1(metastasis associated gene1,MTA1)在乳腺癌浸润与转移中作用。[方法]用逆转录聚合酶链反应(RT-PCR)技术联合ZymedTM免疫组化方法分别检测55例乳腺癌、癌旁组织中MTA1 mRNA和蛋白表达情况。[结果]MTA1 mRNA在乳腺癌中阳性表达率为52.7%(29/55),在癌旁乳腺组织的阳性率23.6%(13/55);两者比较差异有统计学意义(P<0.05)。MTA1蛋白在乳腺癌组织中阳性表达率为49.1%(27/55),在癌旁乳腺组织中阳性表达率为16.4%(9/55);两者比较有统计学差异(P<0.01)。MTA1在乳腺癌组织中的阳性检出率与临床分期、淋巴结转移数目(>4)、病理分级和c-erbB-2阳性相关(P<0.05),与年龄、ER、PR无明显关系(P>0.05)。[结论]MTA1对乳腺癌浸润、转移可能有促进作用,有可能成为预测乳腺癌浸润与转移的参考指标。
[Objective] To investigate the role of metastasis associated gene 1 (MTA1) in infiltration and metastasis of breast cancer. [Method] Reverse transcription polymerase chain reaction (RT-PCR) combined with ZymedTM immunohistochemistry was used to detect the expression of MTA1 mRNA and protein in 55 cases of breast cancer and paracancerous tissues respectively. [Results] The positive rate of MTA1 mRNA in breast cancer was 52.7% (29/55), while the positive rate of MTA1 mRNA in breast cancer was 23.6% (13/55). The difference was statistically significant (P <0.05) . The positive rate of MTA1 protein was 49.1% (27/55) in breast cancer tissues and 16.4% (9/55) in adjacent noncancerous breast tissues. The difference was statistically significant (P <0.01). The positive detection rate of MTA1 in breast cancer was correlated with clinical stage, number of lymph node metastasis (> 4), pathological grade and positive c-erbB-2 (P <0.05), but not with age, ER, > 0.05). [Conclusion] MTA1 may promote infiltration and metastasis of breast cancer and may be a reference index for predicting invasion and metastasis of breast cancer.
