目的:建立HPLC法同时测定人血清中苯巴比妥(PB)、苯妥英(PT)和卡马西平(CBZ)的浓度。方法:分析柱为C18 柱(4.6 mm×250 mm,5μm),流动相为水:甲醇:乙腈(45:45:10),流速为1.0 ml·min-1,检测波长为254 nm,柱温30℃,阿普唑仑为内标物。结果:PB,PT,CBZ的线性范围分别为2.5-80;1.25-40;0.75-24μg·ml-1,最低检测浓度分别为0.2,0.2, 0.1 μg·ml-1,方法回收率分别为99.2%,101.4%,99.4%,日内、日间RSD均小于3%。结论:该方法快速、灵敏,适用于治疗药物监测。
Objective: To establish a HPLC method for the simultaneous determination of phenobarbital (PB), phenytoin (PT) and carbamazepine (CBZ) in human serum. METHODS: The analytical column was C18 column (4.6 mm × 250 mm, 5 μm). The mobile phase consisted of water: methanol: acetonitrile (45:45:10), the flow rate was 1.0 ml · min-1 and the detection wavelength was 254 nm, column temperature 30 ℃, alprazolam as internal standard. Results: The linear ranges of PB, PT and CBZ were 2.5-80, 1.25-40 and 0.75-24μg · ml-1 respectively. The lowest detection concentrations were 0.2, 0.2 and 0.1 μg · ml-1, the recoveries were 99.2%, 101.4% and 99.4%, respectively. The intra-day and inter-day RSD were less than 3%. Conclusion: The method is rapid and sensitive and suitable for the treatment of drug monitoring.
