目的通过室内反选育,获得家蝇对残杀威敏感品系。依据家蝇敏感品系乙酰胆碱酯酶(AChE)敏感度建立不敏感AChE的区分剂量,同时对室外家蝇种群不敏感AChE进行检测,为延缓家蝇抗药性的检测提供依据。方法敏感品系选育采用反汰选法,采用生物化学方法对自然种群中AChE不敏感基因表现型进行检测。结果经过6次反汰选,原敏感品系家蝇对残杀威的敏感度提高到13.87倍。对北京市种群检测结果表明有40%以上个体具有不敏感AChE。结论家蝇敏感品系中含有不敏感AChE,需要继续反选育;自然种群为残杀威抗性种群,应停用残杀威,用其他类型的药剂替代,避免或延缓抗性发展。
Objective Through indoor anti-breeding, access to Musca domestica susceptible strains. Based on the AChE susceptibility of housefly susceptible strains, the discriminating dose of insensitive AChE was established. At the same time, AChE insensitive to outdoor housefly populations was detected, which provided the basis for the delay of the detection of housefly resistance. Methods Sensitive strains were selected by the anti-elimination method, using biochemical methods to detect the natural population AChE insensitive gene phenotype. Results After 6 times of anti-elimination, the susceptibility of Musca domestica to Propoxur increased to 13.87 times. The result of population test in Beijing shows that more than 40% of individuals have insensitive AChE. Conclusion Musca domestica susceptible lines contain insensitive AChE and need to continue anti-selection. Natural population is the predatory population, and should be banned and replaced with other types of agents to avoid or delay the development of resistance.
