目的:研究雷公藤多苷(GTW)对卵巢组织smad4mRNA与GDF-9mRNA表达的影响以及菟丝子黄酮的干预作用。方法:采用4周龄SD雌鼠,据体重随机分3组,分别为空白对照组、GTW组、干预组,连续灌胃12周后处死动物检测指标。结果:GTW组卵巢次级卵泡数值最小,与空白组间差异有统计学意义(P<0.05),与干预组间差异也有统计学意义(P<0.01),空白组与干预组间差异没有统计学意义(P>0.05);黄体数比较各组间差异没有统计学意义(P>0.05)。GTW组smad4mRNA表达与空白组间差异有统计学意义(P<0.01),与干预组间差异也有统计学意义(P<0.05);空白组与干预组间差异没有统计学意义(P>0.05)。GTW组GDF-9mRNA表达与各组比较差异均有统计学意义(P<0.01),空白组GDF-9mRNA表达与干预组间差异没有统计学意义(P>0.05)。结论:雌性幼鼠服用雷公藤多苷可造成卵巢组织病理改变,GTW造成雌性生殖功能损伤的作用位点集中在卵巢局部而不是性腺轴的上游部位,主要损伤的是颗粒细胞和卵母细胞,表现为局部组织中smad4mRNA、GDF-9mRNA表达的下降,尤其是GDF-9mRNA的表达下降对卵泡的发育、分化、成熟具有重要影响。
OBJECTIVE: To study the effects of GTW on the expression of smad4 mRNA and GDF-9 mRNA in ovarian tissue and the intervention of dodder flavonoids. Methods: SD female rats of 4 weeks old were randomly divided into 3 groups according to body weight. They were blank control group, GTW group and intervention group, respectively. After 12 weeks of continuous gavage, animals were sacrificed. Results: The value of ovary secondary follicles in GTW group was the smallest, with a significant difference between the two groups (P <0.05), and the difference between the two groups was also statistically significant (P <0.01). There was no statistical difference between the blank group and the intervention group (P> 0.05). There was no significant difference in the number of corpus luteum between groups (P> 0.05). There was a significant difference between the two groups (P <0.01), the difference between the two groups was also statistically significant (P <0.05), there was no significant difference between the blank group and the intervention group (P> 0.05) . The expression of GDF-9mRNA in GTW group was significantly different from that in each group (P <0.01). There was no significant difference between GDF-9mRNA expression group and intervention group (P> 0.05). CONCLUSION: Tripterygium wilfordii glycosides can induce ovarian histopathological changes in female rats. The site of GTW-induced female reproductive impairment is located in the ovary rather than the upstream part of the gonadal axis. The major damage is granulosa cells and oocytes, The performance of local tissue smad4mRNA, GDF-9mRNA expression decline, especially the decline of GDF-9mRNA on follicular development, differentiation, maturation have a significant impact.
