目的探讨大鼠死后不同时间肝甘油醛3-磷酸脱氢酶(GAPDH)mRNA的降解情况。方法24只大鼠断颈处死后置于20℃温度控制系统内,利用荧光标记逆转录聚合酶链反应(RT-PCR)技术检测大鼠肝GAPDH mRNA在死后即刻至72h的相对表达量的变化情况。结果在死后即刻至48h大鼠肝组织内均可检测到GAPDH mRNA,且其扩增产物呈下降趋势。结论荧光RT-PCR技术检测大鼠肝组织GAPDH mRNA,在死后的降解呈现出一定的规律,可为死亡时间(PM I)推断提供新的指标。
Objective To investigate the degradation of hepatic glyceraldehyde-3-phosphate dehydrogenase (GAPDH) mRNA at different time points after death in rats. Methods Twenty-four rats were sacrificed and then placed in a temperature-controlled system at 20 ℃. The relative expression of GAPDH mRNA in rat liver immediately after the death was measured by RT-PCR (fluorescence-labeled reverse transcription-polymerase chain reaction) Changes. Results GAPDH mRNA was detected in the liver of rats immediately after death to 48 hours, and the products of amplification showed a decreasing trend. Conclusion The detection of GAPDH mRNA by fluorescence RT-PCR in rat liver tissue shows some regularity after the death, which can provide a new index for estimating the death time (PM I).
