目的探讨脂肪酸分析方法在种水平上对嗜肺军团菌的判断能力;将脂肪酸分型方法与传统的血清分型方法进行比较,了解不同血清型嗜肺军团菌株脂肪酸成分的异同。方法选取近3年从广东地区空调冷却塔水中分离到的112株嗜肺军团菌分离株,提取脂肪酸,利用MIDI公司Sherlock系统进行菌体脂肪酸成分分析,使用SPSS 13.0软件对获得的数据资料进行统计分析及聚类分析。结果脂肪酸分析法判定嗜肺军团菌的符合率为84.8%,所有菌株共有的脂肪酸成分有9种,主要脂肪酸成分有4种,分别为16∶0 iso、15∶0 anteiso、16∶1 w7c和14∶0 iso脂肪酸。血清1型菌株与血清6、7型菌株相比,有3种主要脂肪酸含量差异有统计学意义(P<0.05)。结论可以依据菌体脂肪酸分析方法对嗜肺军团菌进行快速鉴定;嗜肺军团菌菌株的脂肪酸成分稳定,但各血清型间脂肪酸含量存在一定差异;MIDI数据库中军团菌脂肪酸数据仍有待完善。本研究同时提供了广东地区空调冷却塔水嗜肺军团菌株脂肪酸组成及分型数据。
OBJECTIVE To investigate the ability of fatty acid assay to judge Legionella pneumophila at the species level. To compare the fatty acid composition of different serogroups Legionella pneumophila strains by comparing fatty acid typing methods with traditional serotyping methods. Methods A total of 112 Legionella pneumophila isolates isolated from the air-conditioner cooling tower water in Guangdong Province in the past three years were selected to extract fatty acids. The fatty acid composition of the bacteria was analyzed by using the Sherlock system of MIDI Company. The obtained data were statistically analyzed by SPSS 13.0 software Analysis and cluster analysis. Results The coincidence rate of Legionella pneumophila determined by fatty acid analysis was 84.8%. There were 9 kinds of fatty acids in all the strains, and the main fatty acids were 4: 16: 0 iso, 15: 0 anteiso, 16: 1 w7c and 14:0 iso fatty acids. There was significant difference in the contents of three major fatty acids between serotype 1 and serotype 6 and 7 strains (P <0.05). Conclusion Legionella pneumophila can be rapidly identified based on the method of bacterial fatty acid analysis. The fatty acid composition of Legionella pneumophila strains is stable, but the fatty acid content of each serotype is different. The data of Legionella fatty acids in the MIDI database still need to be improved. This study also provides data on the fatty acid composition and typing of Legionella pneumophila in air-conditioning cooling towers in Guangdong.
