目的:探讨儿茶酚氧位甲基转移酶(catechol-O-methyltransferase,COMT)的活性与子宫内膜癌发生、发展的关系。方法:建立体外反应体系,以3,4-二羟基苯甲酸(3,4-dihydroxybenzoic acid,DBA)为底物,采用反相高效液相色谱法检测终反应产物4-羟基-3-甲氧基苯甲酸(4-hydroxy-3-methoxybenzoic acid,4-OH-3-MBA)的浓度,以此反映COMT的活性。应用此方法检测子宫内膜癌组织、正常子宫内膜组织(对照组)及待检者外周血中COMT活性,并比较子宫内膜癌组和对照组患者中COMT的活性差异。结果:可溶性COMT(soluble-COMT,S-COMT)是子宫内膜组织中COMT的主要活性形式,子宫内膜组织中S-COMT活性明显高于外周血中S-COMT的活性(P=0.000);子宫内膜癌组织中S-COMT的活性明显低于对照组,差异有统计学意义(P=0.001)。低分化的子宫内膜癌组织中S-COMT的活性明显低于高、中分化的子宫内膜癌组织,差异有统计学意义(P=0.042)。结论:人子宫内膜组织中COMT的活性改变可能与子宫内膜癌的发生、发展有关。
Objective: To investigate the relationship between the activity of catechol-O-methyltransferase (COMT) and the occurrence and development of endometrial carcinoma. Methods: The in vitro reaction system was established. The final reaction products of 4-hydroxy-3-methoxy-4-methoxybenzoic acid The concentration of 4-hydroxy-3-methoxybenzoic acid (4-OH-3-MBA) reflects the activity of COMT. This method was used to detect COMT activity in the endometrial cancer tissue, normal endometrium (control group) and the peripheral blood of the test subjects, and to compare the differences in COMT activity between the endometrial cancer group and the control group. RESULTS: Soluble COMT (S-COMT) was the main active form of COMT in endometrial tissue. The activity of S-COMT in endometrial tissue was significantly higher than that of S-COMT in peripheral blood (P = 0.000) ; The activity of S-COMT in endometrial carcinoma was significantly lower than that in the control group, the difference was statistically significant (P = 0.001). The activity of S-COMT in poorly differentiated endometrial carcinoma was significantly lower than that in high and moderately differentiated endometrial carcinoma with a statistically significant difference (P = 0.042). Conclusion: The changes of COMT activity in human endometrium may be related to the occurrence and development of endometrial cancer.
