目的 为观察不平衡支链氨基酸对胃癌细胞体外增殖和对化疗药物5-FU、MMC和MTX敏感性的影响,为肿瘤的氨基酸失衡疗法提供实验和理论基础。方法 在不含L-Val的MEM培养基基础上,以普通MEM培养基L-Val(46mg/L)和L-Leu含量(52mg/L)为准,配制12种限制L-Val、增加L-Leu的不平衡支链氨基酸培养基,用MTT法观察其对胃癌细胞株SGC-7901体外增殖的影响和对该细胞株化疗药物敏感性的影响。结果 限制L-Val至正常的1/4对 SGC-7901细胞即有明显的抑制作用(p<0.01),随着L-Val浓度的进一步降低,抑制作用更加显著。增加 L-Leu至正常的 4倍,可显著增强 L-Val缺乏对SGC-7901细胞的抑制作用。不平衡支链氨基酸可提高化疗药物5-FU、MMC和MTX对SGC-7901的杀伤作用。结论 不平衡支链氨基酸对胃癌细胞体外增殖有抑制作用,与化疗药物有协同抗肿瘤效应。在肿瘤的氨基酸失衡疗法中,不仅要调整某种氨基酸的绝对量,更重要的是调整该氨基酸与其他氨基酸的比例关系。
Objective To observe the effect of unbalanced branched-chain amino acids on the proliferation of gastric cancer cells and sensitivity to 5-FU, MMC and MTX of chemotherapeutic drugs, and provide experimental and theoretical basis for amino acid imbalance therapy of tumors. Methods Based on the MEM medium without L-Val, L-Val (46 mg/L) and L-Leu (52 mg/L) in common MEM medium were used to prepare 12 kinds of limiting L-Val, increasing L -Leu unbalanced BCAA medium was used to observe the effect of the MTT method on the proliferation of gastric cancer cell line SGC-7901 in vitro and the chemotherapeutic sensitivity of the cell line. RESULTS: Limiting L-Val to 1/4 of the normal SGC-7901 cells significantly inhibited SGC-7901 cells (p<0.01). With the further decrease of L-Val concentration, the inhibitory effect was even more pronounced. Increasing 4-fold L-Leu to normal can significantly enhance the inhibitory effect of L-Val deficiency on SGC-7901 cells. Unbalanced branched-chain amino acids can enhance the killing effect of chemotherapeutic agents 5-FU, MMC, and MTX on SGC-7901. Conclusion Imbalanced branched-chain amino acids can inhibit the proliferation of gastric cancer cells in vitro and synergistic anti-tumor effects with chemotherapy drugs. In the amino acid imbalance therapy of tumors, not only the absolute amount of an amino acid needs to be adjusted, but more importantly, the proportional relationship between the amino acid and other amino acids is adjusted.
