目的初步探讨不同浓度的五味子乙素(Sch B)对人神经母细胞瘤SH-SY5Y细胞及C6胶质瘤细胞生长及增殖的影响。方法体外培养人神经母细胞瘤SH-SY5Y细胞及C6胶质瘤细胞,分别以1~13μmol/L Sch B作用于SH-SY5Y细胞及C6细胞48 h。倒置显微镜下观察两种肿瘤细胞的形态学及数量变化,四唑盐比色法(MTT法)观察不同浓度的Sch B对细胞生长增殖的影响,酶标仪检测各组吸光度。结果与对照组相比,9~13μmol/L Sch B作用48 h均可引起SH-SY5Y细胞生长增殖抑制,1~7μmol/L Sch B对SH-SY5Y细胞没有毒性作用;而5μmol/L Sch B即可对C6胶质瘤细胞的生长增殖产生抑制作用。结论 Sch B对SH-SY5Y细胞及C6胶质瘤细胞的生长增殖均有明显的抑制作用,且C6胶质瘤细胞对Sch B的增殖抑制作用更为敏感。
Objective To investigate the effects of different concentrations of schisandrin B on the growth and proliferation of human neuroblastoma SH-SY5Y cells and C6 glioma cells. Methods Human neuroblastoma SH-SY5Y cells and C6 glioma cells were cultured in vitro. SH-SY5Y cells and C6 cells were treated with 1 ~ 13μmol / L Sch B for 48 h. Morphological and quantitative changes of the two tumor cells were observed under an inverted microscope. The effects of Sch B at different concentrations on the cell proliferation were observed by MTT method. The absorbance of each group was detected by microplate reader. Results Compared with the control group, the Schβ5 cells treated with 9 ~ 13μmol / L SchB for 48 h all could inhibit the proliferation of SH-SY5Y cells. Sch B 1 ~ 7μmol / L showed no toxic effect on SH-SY5Y cells. Can inhibit the growth and proliferation of C6 glioma cells. Conclusion Sch B significantly inhibits the growth and proliferation of SH-SY5Y cells and C6 glioma cells, and C6 glioma cells are more sensitive to the inhibition of Sch B proliferation.
