目的:建立HPLC同时测定决明子中红镰霉素龙胆二糖苷、决明子苷、决明子苷C含量的方法。方法:采用HPLC,Dionex Acclaim C18柱(4.6 mm×250 mm,5μm),流动相乙腈-四氢呋喃-1%冰醋酸(17∶2∶81),流速1 mL·min-1,柱温30℃,检测波长278 nm。结果:红镰霉素龙胆二糖苷、决明子苷、决明子苷C的进样量分别在0.218~1.090,0.188~0.940,0.200~1.000μg和峰面积呈良好的线性关系,回归方程分别为Y=54.017X-0.344 6(r=0.999 9),Y=78.63X-0.354(r=0.999 9),Y=74.098X-0.433 5(r=0.999 9);平均加样回收率分别为98.43%(RSD 1.75%),101.89%(RSD 1.10%),102.46%(RSD 1.15%)。结论:该方法快速、灵敏、准确、可靠、重复性好,可用于中药决明子药材的质量控制。
OBJECTIVE: To establish a method for the simultaneous determination of erythromycins gentiobioside, cassia seed and cassia glycoside C in cassia seed by HPLC. Methods: HPLC, Dionex Acclaim C18 column (4.6 mm × 250 mm, 5 μm), mobile phase acetonitrile-tetrahydrofuran-1% glacial acetic acid (17:2:81), flow rate 1 mL · min- Detection wavelength of 278 nm. Results: The injection rates of erythromycins gentiobioside, cassia seed and cassia seed C were 0.218-1.090,0.188-0.940, 0.200-1.000μg and the peak areas were linear. The regression equations were Y = (R = 0.999 9), Y = 78.63X-0.354 (r = 0.999 9) and Y = 74.098X-0.433 5 (r = 0.999 9) respectively. The average recoveries were respectively 98.43% (RSD 1.75%), 101.89% (RSD 1.10%), 102.46% (RSD 1.15%). Conclusion: The method is rapid, sensitive, accurate, reliable and reproducible. It can be used for the quality control of Chinese herbal medicine Cassia.
