目的:观察西洋参茎叶20s-原人参二醇组皂苷(PQDS)对大鼠实验性脑缺血的保护作用。方法:采用大鼠结扎双侧颈总动脉伴低血压建立急性不完全性脑缺血模型,计算脑指数及脑含水量,测定血清超氧化物歧化酶(SOD)、一氧化氮合酶(NOS)活力及血清丙二醛(MDA)、一氧化氮(NO)含量,同时测定脑组织SOD、钠-钾-ATP酶(Na+-K+-ATPase)、钙-镁-ATP酶(Ca2+-Mg2+-ATPase)活力及MDA、钙离子(Ca2+)、乳酸(LA)含量。结果:PQDS可明显降低脑指数及脑含水量,亦能明显降低缺血后血清和脑组织MDA,Ca2+,LA,NO含量及NOS活性,提高SOD,Na+-K+-ATPase和Ca2+-Mg2+-ATPase活力。结论:PQDS对大鼠实验性脑缺血具有明显保护作用,其机制可能与抑制脂质过氧化物反应,提高抗氧化酶活性,抑制NO生成,降低LA酸中毒和细胞内钙超载,改善细胞能量代谢等作用有关。
Objective: To observe the protective effect of 20s-protopanaxadiol Saponins (PQDS) on the experimental cerebral ischemia in rats. Methods: Acute incomplete cerebral ischemia model was established by ligating bilateral common carotid arteries with hypotension in rats. Brain index and brain water content were calculated. Serum levels of superoxide dismutase (SOD), nitric oxide synthase (NOS) ) Activity and serum malondialdehyde (MDA) and nitric oxide (NO) contents were determined. The activities of SOD, Na + -K + -ATPase and Ca2 + -Mg2 + ATPase activity and MDA, calcium (Ca2 +), lactate (LA) content. Results: PQDS significantly reduced brain index and brain water content, but also significantly decreased MDA, Ca2 +, LA, NO content and NOS activity in serum and brain tissue, increased SOD, Na + -K + -ATPase and Ca2 + -Mg2 + -ATPase vitality. CONCLUSION: PQDS has a significant protective effect on experimental cerebral ischemia in rats. The mechanism may be related to inhibition of lipid peroxidation, increase of antioxidant enzyme activity, inhibition of NO production, decrease of LA acidosis and intracellular calcium overload and improvement of cells Energy metabolism and other related.
