目的 :研究P38信号通路 (P38mitogen activatedproteinki nase ,P38MAPK)在人脐静脉内皮细胞表达P 选择素以及糖尿病动脉粥样硬化中的作用。方法 :分别以高葡萄糖、糖基化终产物 (AGE)、高胰岛素和过氧化氢刺激人脐静脉内皮细胞系ECV 30 4 ,再以P38MAPK特异性抑制剂SB2 0 35 80预处理 ,观察ECV 30 4细胞系中磷酸化P38MAPK和P 选择素的表达。结果 :4种刺激因素均可单独激活P38MAPK ,使磷酸化P38MAPK表达量明显增加 ,P 选择素表达量也明显增加 ;以SB2 0 35 80预处理后 ,P 选择素的表达被明显抑制。结论 :P38MAPK是P 选择素的上游信号分子 ,也可能是动脉粥样硬化发生的始动信号之一
AIM: To investigate the role of P38 mitogen activated protein kinase (P38MAPK) in the expression of P-selectin and atherosclerosis in human umbilical vein endothelial cells. Methods: The human umbilical vein endothelial cell line ECV 30 4 was stimulated with high glucose, AGE, high insulin and hydrogen peroxide respectively, and then pretreated with P38MAPK specific inhibitor SB2 0 35 80. ECV 30 4 cell lines phosphorylated P38MAPK and P-selectin expression. Results: P38MAPK could be activated by all the four stimuli. The expression of phosphorylated P38MAPK was significantly increased and the expression of P-selectin was significantly increased. The expression of P-selectin was significantly inhibited by SB203580 pretreatment. Conclusion: P38MAPK is the upstream signal molecule of P-selectin and may also be one of the initiating signals of atherosclerosis
